Antibodies to the Glutamate Receptors

Monoclonal Antibody against Kainate receptors:
MAb 4F5 was raised against a portion of the putative extracellular domain of GluR5 receptor subunit protein (aa 233-518) using a trpE bacterial expression system. This antibody binds only to receptors belonging to the kainate class, e.g. , GluR 5, 6, and 7 and not to GluR 1, 2, 3, and 4. MAb 4F5 has been used to study distribution of kainate receptor subunits in primate cortex. MAb 4F5 reacts with a 110 kDa protein in crude membrane preparations of cerebral cortex, cerebellum, and hippocampus. 4F5 is an excellent antibody for immunohistochemistry in rat, monkey, and human tissues.

Monoclonal antibody against GluR2 and GluR4:
MAb 3A11 binds specifically to GluR2 and GluR4 of the AMPA class subunits (GluR1-4). It was prepared against a fusion protein from the N-terminal, putative extracellular portion of GluR2. This is the only known mAb that differentiates GluR2 from GluR3.

Monoclonal antibody against NMDAR1:
MAb 54.1 was raised against bacterial trpE fusion protein corresponding to NMDAR1 residues 660-811 (representing the putative intracellular loop between transmembrane regions III and IV). This antibody is an excellent reagent for immunolocalization of NMDAR1 receptors in brain.

Monoclonal antibodies against mGluR1a
MAb G209-488 and G209-2048 recognize rat mGluR1a. They does not cross-react with the other splice mGluR1 variants, mGluR1b, and mGluR1c. Additionally G209-488 and G209-2048 do not cross-react with mGluR5, the most closely related mGluR family member. These antibodies were characterized by western blot analysis using rat brain membranes, and by immunohistochemical analysis using frozen rat brain tissue sections. A GST fusion protein containing sequences from the C-terminus of mGluR1a was used as immunogen.
