BACKGROUND
The JJ319 antibody reacts with the rat CD28 molecule, a potent costimulatory receptor for T cells. In the periphery, CD28 is expressed on virtually all T cells bearing alpha beta TCR, on most gamma delta TCR-bearing T cells, and on a subset of NK cells. In the thymus, CD28 expression is regulated during the maturation of alpha beta TCR-bearing T cells. The major population of CD4+CD8+ TCRlow thymocytes are CD28- to CD28low, while the small subset of CD4+8+CD3intermediate thymocytes express the highest levels of CD28. CD4-CD8- thymocytes include CD28- and CD28+ subsets, and almost all CD4+CD8-and CD4-CD8+ thymocytes are CD28+. Soluble JJ319 mAb costimulates both proliferation and IL-2 production of purified T cells activated by anti-TCR mAb.1
USAGE
This antibody has been tested by LAL assay for endotoxin level and by immunofluorescent staining (lesser than or equal to 1 µg/million cells) with flow cytometric analysis to assure specificity and reactivity. Other reported applications include immunoprecipitation and functional studies. Since applications vary, each investigator must determine dilutions appropriate for individual use.
