Dear David,
It sounds like you have put alot of time in this and I have seen
the same thing with other markers. Have you tried to add pure bcl-2 or an
extract that has bcl-2 to your antibody prior to adding it to the cells.
If you still get the same rel. fl. value than I would be leary of the flow
cytometry results. You might also try a low concentration (sub saturation
) of antibody and see what happens. Have you looked at the cells after
they are put through the flow and do they still show the same fl. intensity
in a microscope?
