Autofluorescence and nodes

I am a very new flow operator and would appreciate any comments that =
might help me resolve my problem. I recently had to analyse a piece of =
tumour tissue by flow. My results suggest that the sample showed =
autofluorescence. The original specimen was received in RPMI. From =
this a cell suspension was prepared in PBS containing BSA and sodium =
azide. After washing a number of times, the cells were stained, =
incubated and then analysed. =20
I would appreciate any thoughts about my technique and/or the avoidance =
of this problem in the future. =20
