Greetings Folks,
I have a customer interested in molar limits of detection for green
fluorescent protein, preferably the S65T mutant. I recall earlier
discussions on things such as utility (or lack of) in single copy gene
expression etc.
But does anyone have an idea of the relative fluorescence of GFP (488ex
/535em) to ,say, fluorescein on a per molecule basis (pH 7, aqueous
environment)?
I have found a reference for quantum yield of the native molecule (0.78
...Morise et al. 1974. Biochemistry, 13: 2656-2662).
I would appreciate your input and will post a summary.
