SPECIFICITY
The 17A2 antibody reacts with the T-cell receptor-associated CD3 complex, which is expressed on thymocytes and mature T lymphocytes of all mouse strains tested.(1) Plate-bound mAb 17A2 can induce IL-2 production by T cells in the absence of accessory cells.(1) Binding of 17A2 antibody to T cells can be blocked by the anti-CD3e mAb 145-2C11, suggesting that 17A2 antibody recognizes an epitope of the CD3 epsilon chain.(1)
USAGE
The antibody has been tested by immunofluorescent staining (< 1 µg/million cells) with flow cytometric analysis to assure specificity and reactivity. Other reported applications include immunoprecipitation,(1) complement-mediated cytotoxicity,(1,2) immunohistochemical staining of acetone-fixed frozen sections,(1) and in vitro stimulation of T cells. The hamster anti-mouse CD3e mAb 145-2C11 (Cat. no. 01080D) has been reported to induce greater functional responses than 17A2 antibody in assays which depend on presentation of the monoclonal antibody by Fc receptor-positive macrophages.(1) Since applications vary, each investigator must determine dilutions appropriate for individual use.
