

Apoptosis, a term that describes regulated cell death, is a fundamental feature of many biological processes including normal development, homeostasis, and disease. Changes in the plasma membrane of the cell surface are one of the earliest features of cells undergoing apoptosis. In apoptotic cells, the membrane phospholipid phosphatidylserine (PS) is translocated from the inner leaflet of the plasma membrane to the outer leaflet, thereby exposing PS to the external environment. Annexin V is a Ca2+-dependent phospholipid-binding protein that has a high affinity for PS, and is useful for identifying apoptotic cells with exposed PS. Annexin V-FITC can be used in a bivariate analysis with Propidium Iodide (PI) to distinguish between cells undergoing apoptosis (PI negative) and those that are necrotic or dead (PI positive). Since externalization of PS occurs earlier than nuclear changes associated with apoptosis, Annexin V-FITC can identify cells in an earlier stage of apoptosis than assays based on DNA fragmentation. PharMingen's Annexin V-FITC is available in 100 and 200 test sizes and comes with a complete protocol.
