SPECIFICITY
The APA5 antibody reacts with Platelet-Derived Growth Factor (PDGF) Receptor alpha chain (CD140, PDGFR-alpha), a receptor tyrosine kinase which is widely expressed on cells of mesenchymal origin in the embryo and adult and on several other cell types during embryonic development, but not on hematopoietic cells.(1,2) PDGFR-alpha binds to PDGF A and B chains, in contrast to PDGFR-beta, which binds only to the PDGF B chain.(3) Biologically active PDGF is a disulphide-linked dimer, forming the AA, AB, and BB isoforms. Binding to PDGF Receptor induces the formation of receptor dimers (alpha alpha, alpha beta, or beta beta), autophosphorylation, and internalization.(3) The mAb APA5 has been demonstrated to block binding of PDGF-AA to PDGFR-alpha-expressing cells in vitro (1) and to block some PDGF-mediated developmental events in vivo.(1,2)
USAGE
This antibody has been tested by immunofluorescent staining (< 1 µg/million cells) with flow cytometric analysis to assure specificity and reactivity. Other reported applications include western blot analysis,(1) ELISA,(1) immunohistochemical staining of acetone-fixed frozen sections(4) and polyester wax-embedded sections,(1) inhibition of ligand binding,(1) and in vivo blocking of PDGF action.(1,2) Since applications vary, each investigator must determine dilutions appropriate for individual use.
