New Glutamate Receptor Antibodies

PharMingen has released several new antibodies against the glutamate receptor:

Polyclonal Anti-GluR1, Cat. No. 60646N: The antibodies recognize human, rat, monkey, gerbil and hamster GluR1. The GluR1 antibodies do not cross-react with GluR2, GluR3, or GluR4. A synthetic peptide corresponding to the carboxyl terminus (amino acids 877-889: SHSSGMPLGATGL) of rat GluR1 was used as immunogen.

Applications include western blot analysis (0.5­1 µg/ml), immunoprecipitation, and immunohistochemistry of paraformaldehyde- and paraformaldehyde/glutaraldehyde-fixed frozen tissue sections (1­2 µg/ml) for use in light and electron microscopy. In western blots, the antibodies recognize a major band at ~108 kD corresponding to GluR1. Higher molecular weight bands at around 200 kD may represent incompletely reduced complexes of glutamate receptors.


Purified anti-GluR2, Cat. No. 60671A: Monoclonal antibody (mAb) 6C4 recognizes rat, macaque monkey, and dog GluR2. It does not cross-react with GluRs 1,3, 4c, 5, 6, or 7. A recombinant trpE fusion protein containing the putative N-terminal portion (amino acids 175-430) of GluR2 was used as immunogen. In western blot analysis of tissue homogenates, mAb 6C4 recognizes GluR2 as an ~102 kD protein. Other proteins noted by Western blot at 66 kD and lower molecular weights appear to be breakdown products of GluR2.

Applications include western blot analysis (2 µg/ml), immunohistochemistry of 4% paraformaldehyde-fixed frozen tissue sections (2 µg/ml), immunohistochemistry of 4% paraformaldehyde-fixed tissue cultured cells, post embedding immunogold electron microscopy, and radioimmunoassay.


Polyclonal anti-GluR2, Cat. No. 60686N: The antibodies recognize rat GluR2. The GluR2 antibodies do not cross-react with other ionotropic glutamate receptors including GluR1, GluR3, GluR4, GluR4c, GluR5, GluR6, GluR7, KA2, NR1, NR2A, NR2B, NR2C, NR2D, delta 1 or delta 2. The immunogen was a synthetic peptide (VAKNPQNINPSSSQNS), corresponding to amino acid residues 827-842, 20 residues from the C-terminus of rat GluR2.

Applications include western blot analysis (0.5­1.0 µg/ml), and immunohistochemistry of paraformaldehyde- and paraformaldehyde/glutaraldehyde-fixed frozen tissue sections (1­2 µg/ml) for use in light and electron microscopy. In western blots, the antibodies recognize a major band at ~108 kD corresponding to GluR1. Higher molecular weight bands at around 200 kD may represent incompletely reduced complexes of glutamate receptors.


Polyclonal anti-GluR2 and GluR3, Cat. No. 60656N: The antibodies recognize human, rat, monkey, gerbil and hamster GluR2 and GluR3 The antibodies also recognize the GluR4 variant, GluR4c. The GluR2 and GluR3 antibodies do not cross-react with GluR1. With regards to GluR4, they only recognize the GluR4c variant and not GluR4. A synthetic peptide corresponding to the carboxyl terminus (amino acids 850-862: EGYNVYGIESVKI) of rat GluR2 was used as immunogen.

Applications include western blot analysis (0.15 µg/ml), immunoprecipitation, and immunohistochemistry of paraformaldehyde- and paraformaldehyde/glutaraldehyde-fixed frozen tissue sections (1­2 µg/ml) for use in light and electron microscopy. In western blots, the antibodies recognize two poorly separated bands at ~104-108 kD corresponding to GluR2 and GluR3. If present at high at detectable quantities, GluR4c is also detectable by western blot analysis at ~105 kD. Higher molecular weight bands at around 200 kD may represent incompletely reduced complexes of glutamate receptors.


Polyclonal anti-GluR4, Cat. No. 60666N: The antibodies recognize human, rat, monkey, gerbil and hamster GluR4. The GluR4 antibodies do not cross-react with GluR1, GluR2, or GluR3, or GluR4c. A synthetic peptide corresponding to the carboxyl terminus (amino acids 868-881: RQSSGLAVIASDLP) of rat GluR4 was used as immunogen. The peptide sequence is specific to the GluR4 subunit, it is not shared among other GluR subunits.

Applications include western blot analysis (0.5­1.0 µg/ml), immunoprecipitation, and immunohistochemistry of paraformaldehyde- and paraformaldehyde/glutaraldehyde-fixed frozen tissue sections (1­2 µg/ml) for use in light and electron microscopy. In western blots, the antibodies recognize a major band at ~108 kD corresponding to GluR4. Higher molecular weight bands at around 200 kD may represent incompletely reduced complexes of glutamate receptors.
