

SPECIFICITY
The F23.1 antibody reacts with the Vbeta 8.1, Vbeta 8.2, and Vbeta 8.3 T-cell receptors (TCR) of mice having the Vbeta b haplotype (e.g., A, AKR, BALB/c, CBA/Ca, CBA/J, C3H/He, C57BL, C58, DBA/1, DBA/2).(1,2) These gene loci are deleted in Vbeta a (e.g., C57BR, C57L, SJL, SWR) and Vbeta c (e.g., RIII) mice.(2) Vbeta 8.1 TCR-bearing T cells are clonally eliminated in mice expressing superantigen coded by Mtv-7 (Mls-1a, Mlsa) provirus (e.g., AKR, CBA/J, C58, DBA/2), and activation or elimination of Vbeta 8.1 TCR-expressing T cells by this determinant is partially dependent upon presentation by I-E.(3,4) Mtv-43 and/or exogenous MMTV-SW superantigens also cause incomplete elimination of Vbeta 8.1 TCR-bearing T cells.(4) Soluble and plate-bound F23.(1) antibody activates Vbeta 8 TCR-bearing T cells,(1) soluble antibody blocks cytolysis mediated by Vbeta 8 TCR-bearing CTL's,(1) and in vivo treatment of neonatal mice can arrest intrathymic maturation of Vbeta 8 TCR-bearing T cells.(5)
USAGE
This antibody has been tested by immunofluorescent staining (approx. 1µg/million cells) with flow cytometric analysis to assure specificity and reactivity. For flow cytometry of cell suspensions from peripheral lymphoid tissues, it is recommended that multicolor staining be performed to distinguish T lymphocytes from non-T-cells. Other reported applications include immunoprecipitation,(1,6) western blot analysis,(6) in vitro stimulation of T cells expressing Vbeta 8.1 or 8.2 TCR,(1) in vitro blocking of cytolysis,(1) and in vivo inhibition of thymocyte maturation.(5) Since applications vary, each investigator must determine dilutions appropriate for individual use.
